"Your body is fighting itself". Yes. That is exactly why multiple gene-splices are so hard to make stick. It's not just your cells though. The very DNA you want to be changing rejects itself, calls itself faulty or bad. Now, whether it actually is faulty or bad is up to debate of course. As long as you don't have a natural, extremely high regeneration property, you will most likely die when you receive your splices. When he uses his "healing" elixir, he is forcing a toxic substance into his body that has to stop fighting itself long enough to fight it.
It's like Blade! You've got a treatment against your condition, but you have to constantly up the dosage and find a stronger drug in order to keep yourself alive. But he has to be extremely careful with it, because he is injecting himself with deadly poisons and diseases. It's not like he can just give himself cancer or AIDS, those would just work with his already fucked up genetics to make him die faster. He has to be constantly poisoned in order to function properly. If he wasn't an extremely important toxicologist, he wouldn't have had access to the poisons he is using to keep himself alive with, and he'd already be dead.
